Detection and quantification of endocannabinoids and related lipids by LC-MS/MS
Endocannabinoids (ECs) are synthesized from membrane phospholipids precursors. Initiation of biosynthesis can be stimulated by a rise in intracellular Ca2+, or other triggers such as the activation of metabotropic neurotransmitter receptors. These stimuli thereby elevate EC synthesis "on-demand" above basal levels. By determining EC levels and related lipids, we can thus assess the activity status of the EC system. In this Research Unit, the hypothesis is put forward that the EC system is involved in homeostatic processes and that in pathological states, overactivation or underactivity of the EC system contributes to allostatic conditions.
Determination of ECs and EC metabolites requires highly sensitive and accurate analytical methods. These requirements are met by today’s state-of-the -art LC-MS/MS technology.
